Due to their various ranges of sensitivity, types of fluorophores, readout modes, and ease of operation, fluorescence-based assays are widely used in high-throughput screening in a broad range of activities in life-science research, e.g. such as molecular dynamics and interactions, enzymatic activities, signal transduction, cell viability, and quantification of molecules, organelles, or cells.

96-Well Plate Format E. coli Growth

The functionality of genetic fluorescence-based systems can be measured by a multi-mode 96-well microplate reader under various growth/incubation optimal/non-optimal conditions. Both average fluorescence intensities from various modes and average cell biomass (measured by optical densities) of each individual sample are monitored from cells at certain sampling intervals and periods.

E. coli Growth Fluorescence Microscope Assay

Single-cell data can be visualized by a set of epi-fluorescence light source, and a filter cube containing an excitation filter, a dichroic mirror, and an emission filter of a specific fluorescence mode using an inverted microscope. Along with fluorescence images, phase contrast images of the same location can also be captured by a CCD camera for concomitant cell morphological analysis. Microscopy images are captured using MicroManager, open-source software tool, and analyzed using customized scripts in MATLAB.

The addition of glycerol stabilizes the frozen bacteria, preventing damage to the cell membranes and keeping the cells alive for long term storage. DAMP lab offers up to 2 years of storage time for glycerol stocks of Bacteria cell culture.

The DAMP Lab currently provides mRNA enrichment for both bacterial RNA (rRNA removal) and mammalian RNA (poly-A selection) inputs, cDNA library preparation using the NEB Ultra kit, indexing and quality control before performing high-throughput sequencing via Illumina HiSeq 2000.
